DEPOTONE

*Dysfunctional uterine bleeding:* ORAL: Adult (female): 2.5-10 mg daily for 5-10 days, repeated for 2 cycles, begin treatment on day 16-21 of cycle. *Secondary amenorrhoea:* ORAL: Adult (female): 2.5-10 mg daily for 5-10 days, repeated for 3 cycles, begin treatment on day 16-21 of cycle. *Mild to moderate endometriosis:* ORAL: Adult (female): 10 mg 3 times a day for 90 consecutive days, begin treatment on day 1 of cycle. BY IM INJ: Adults: 50 mg each week or 100 mg every 2 week for 6 months or longer. *Progestogenic opposition of oestrogen HRT:* ORAL: Adult (female): 10 mg daily for the last 14 days of each 28-day oestrogen HRT cycle. *Breast carcinoma:* ORAL: Adult: 0.4-1.5 gm daily, dose adjusted according to response (by specialist only). *Endometrial and renal carcinoma:* ORAL: Adult: 200-600 mg daily, dose adjusted according to response (by specialist only). *Prostatic carcinoma:* ORAL: Adult: 100-600 mg daily, dose adjusted according to response (by specialist only). *Contraception:* BY DEEP IM INJ: Females of childbearing potential: 150 mg every12 weeks, to be administered within the first 5 days of cycle or within first 5 days after parturition (delay until 6 weeks after parturition if breast-feeding). BY SC INJ: Females of childbearing potential: 104 mg, administered within first 5 days of cycle or within 5 days postpartum (delay until 6 weeks postpartum if breast-feeding), injected into anterior thigh or abdomen, dose only suitable if no hormonal contraceptive use in previous month.

Administer IM only. Shake container vigorously for at least 1 min before giving injection. Inject slowly, over 5-7 sec, into deltoid muscle. Do not inject into gluteal muscle. Pat site lightly after injection. Do not rub it.

Hormone dependent carcinoma, pregnancy, acute porphyrias, severe arterial disease, undiagnosed vaginal bleeding, history of breast cancer but can be used after 5 years if no evidence of disease and non-hormonal contraceptive methods unacceptable

Severe depression, diabetes. While using as contraceptive wait 6 weeks post-partum if lactating, risk of breast cancer, history during pregnancy in disturbances of lipid metabolism or deterioration of otosclerosis or pruritus

Breast discomfort, depression, dizziness, headache, indigestion, loss of vision during treatment (discontinue treatment if papill oedema or retinal vascular lesions), menstrual irregularities, nausea, pruritus, vomiting, weight gain, disturbance ofappetite, injection site-reactions, reduced bone mineral density, skin disorders. Rare: Osteoporosis, osteoporotic fractures
